چکیده

To address the unsolved question of PSD-95 function I used short hairpin RNA (shRNA) to knock-down protein expression. shRNA mediated knock-down of protein expression is one of the established methods exploiting the naturally occurring expression control mechanism known as RNA interference (RNAi). Cells express shRNAs that are complementary to the mRNA they control. The shRNAs are processed and bound by two complexes called DICER (***) and RISC. Subsequentially the complementary the RISC complex binds the mRNA and the mRNA is degraded. This mRNA depletion decreases the protein level. In shRNA experiments, the short hairpin RNAs are expressed from a DNA plasmid vector. Polymerase III promoters are generally used as they reliably terminate transcription. shRNA constructs affect the targeted mRNA with variable efficiency. I generated four potential shRNA constructs to suppress PSD-95 expression. To test for efficiency I co-expressed the shRNA together with PSD-95 cDNA in HEK293T cells. I used the shRNA vector without a hairpin insert as a control to visualize unaffected PSD-95 expression.

Cyclin-dependent kinase 5 phosphorylates the N-terminal domain of the postsynaptic density protein PSD-95 in neurons.

PSD-95 (postsynaptic density 95) is a postsynaptic scaffolding protein that links NMDA receptors to the cytoskeleton and signaling molecules. The N-terminal domain of PSD-95 is involved in the synaptic targeting and clustering of PSD-95 and in the clustering of NMDA receptors at synapses. The N-terminal domain of PSD-95 contains three consensus phosphorylation sites for cyclin-dependent kinase ...

Activating transcription factor 4 (ATF4) modulates post-synaptic development and dendritic spine morphology

The ubiquitously expressed activating transcription factor 4 (ATF4) has been variably reported to either promote or inhibit neuronal plasticity and memory. However, the potential cellular bases for these and other actions of ATF4 in brain are not well-defined. In this report, we focus on ATF4's role in post-synaptic synapse development and dendritic spine morphology. shRNA-mediated silencing of...

Acute Inactivation of PSD-95 Destabilizes AMPA Receptors at Hippocampal Synapses

Postsynatptic density protein (PSD-95) is a 95 kDa scaffolding protein that assembles signaling complexes at synapses. Over-expression of PSD-95 in primary hippocampal neurons selectively increases synaptic localization of AMPA receptors; however, mice lacking PSD-95 display grossly normal glutamatergic transmission in hippocampus. To further study the scaffolding role of PSD-95 at excitatory s...

A proteasome-sensitive connection between PSD-95 and GluR1 endocytosis.

Synaptic transmission at excitatory synapses can be regulated by changing the number of synaptic glutamate receptors (GluRs) through endocytosis and exocytosis. The endocytosis of GluRs has recently been shown to require the activity of the ubiquitin-proteasome system (UPS): proteasome inhibitors or dominant negative forms of ubiquitin block the ligand-stimulated internalization of GluRs. We ha...

Effect of disrupting N-methyl-d-aspartate receptor-postsynaptic density protein-95 interactions on the threshold for halothane anesthesia in mice.

BACKGROUND The authors' previous studies have shown that clinically relevant concentrations of inhalational anesthetics dose-dependently and specifically inhibit the PSD-95, Dlg, and ZO-1 (PDZ) domain-mediated protein interactions between postsynaptic density protein 95 (PSD-95) and N-methyl-d-aspartate receptors, and that the knockdown of spinal PSD-95 by intrathecal injection of PSD-95 antise...
